Total Knee Replacements are increasingly being performed on younger, and more active, people for whom the rate of revision due to wear is more than double that of traditional populations. The proposed study will utilize a state-of-the-art mechanical knee simulator to examine the effects of prosthesis malalignment and high demand activity on wear. Wear dynamics will be assessed using a novel system of highly sensitive radioisotope labelling. Bioactivity of the wear debris will be evaluated via a novel assay system. The resultant knowledge will potentially increase the longevity of total knee prostheses and reverse the trend of increasing revision rates.
